Pharmacological action – D-vitamin-like, antipsoriatic, glucocorticoid, local anti-inflammatory.
Pharmacodynamics
Calcipotriol is a synthetic analog of the active metabolite of vitamin D. Causes inhibition of keratinocyte proliferation and accelerates their morphological differentiation.
Betamethasone – GCS for external use; has a local anti-inflammatory, antipruritic, vasoconstrictor and immunosuppressive effect.
Pharmacokinetics
With external application of Daivobet, the absorption of calcipotriol and betamethasone through intact skin is less than 1%. When the drug is applied to psoriatic plaques and under occlusive dressings, the absorption of betamethasone increases. Since a depot of the drug is created in the skin, its elimination from the skin occurs within a few days. Betamethasone is metabolized in the liver and kidneys with the formation of glucuronides and sulfoesters, excretion is through the intestines and kidneys.
Indications
Chronic vulgar plaque psoriasis.
Contraindications
hypersensitivity to any of the components of the drug;
diseases accompanied by impaired calcium metabolism;
severe renal and hepatic insufficiency;
viral (including herpes or chicken pox), fungal, bacterial and parasitic infections of the skin, rosacea, rosacea, acne vulgaris, perioral dermatitis, skin manifestations of tuberculosis and syphilis, skin atrophy, skin ulcers, increased fragility of skin vessels, ichthyosis, perianal and genital itching;
psoriatic erythroderma, guttate, exfoliative, pustular psoriasis;
children under 18 years of age (there is no experience with Daivobet®).

Calcipotriol ist ein synthetisches Analogon des aktiven Metaboliten von Vitamin D. Verursacht eine Hemmung der Keratinozytenproliferation und beschleunigt ihre morphologische Differenzierung.
Betamethason – GCS zur au?erlichen Anwendung; wirkt lokal entzundungshemmend, juckreizstillend, vasokonstriktorisch und immunsuppressiv.
Pharmakokinetik
Bei au?erlicher Anwendung von Daivobet betragt die Resorption von Calcipotriol und Betamethason durch die intakte Haut weniger als 1 %. Wenn das Medikament auf Psoriasis-Plaques und unter Okklusivverbanden aufgetragen wird, erhoht sich die Resorption von Betamethason. Da ein Depot des Arzneimittels in der Haut gebildet wird, erfolgt dessen Ausscheidung aus der Haut innerhalb weniger Tage. Betamethason wird in Leber und Nieren unter Bildung von Glucuroniden und Sulfoestern metabolisiert, die Ausscheidung erfolgt uber Darm und Nieren.
Hinweise
Chronische vulgare Plaque-Psoriasis.
Kontraindikationen
Uberempfindlichkeit gegen einen der Bestandteile des Arzneimittels;
Krankheiten, die mit einem gestorten Calciumstoffwechsel einhergehen;
schwere Nieren- und Leberinsuffizienz;
virale (einschlie?lich Herpes oder Windpocken), Pilz-, bakterielle und parasitare Infektionen der Haut, Rosacea, Rosacea, Akne vulgaris, periorale Dermatitis, Hautmanifestationen von Tuberkulose und Syphilis, Hautatrophie, Hautgeschwure, erhohte Zerbrechlichkeit der Hautgefa?e, Ichthyose, perianaler und genitaler Juckreiz;
psoriatische Erythrodermie, guttata, exfoliative, pustulose Psoriasis;
Kinder unter 18 Jahren (es liegen keine Erfahrungen mit Daivobet® vor).
