Tarceva (Erlotinib) 100mg, 30

Indications

locally advanced or metastatic non-small cell lung cancer after failure of one or more chemotherapy regimens;

first-line therapy for locally advanced, unresectable or metastatic pancreatic cancer in combination with gemcitabine.

Contraindications

severe hypersensitivity to erlotinib or any component of the drug;

pregnancy and breastfeeding.

Carefully:

liver dysfunction;

age up to 18 years.

Side effects

Side effects were distributed by frequency as follows: very often (≥1/10); often (≥1/100, <1/10); infrequently (≥1/1000, <1/100); rarely (≥1/10000, <1/1000); very rarely (<1/10000), including isolated cases.

Monotherapy for non-small cell lung cancer

The most common adverse events, regardless of the relationship with the drug: rash (75%) and diarrhea (54%), most of which are grade 1 and 2 and do not require medical intervention. Rash and grade 3–4 diarrhea occurred in 9% and 6% of patients with non-small cell lung cancer, each of which required discontinuation of therapy in 1% of patients and dose adjustment of erlotinib in 6% and 1% of patients, respectively. The average time period before the onset of a rash is 8 days, before the onset of diarrhea is 12 days.

The most common side effects:

From the gastrointestinal tract: anorexia, diarrhea, nausea, vomiting, stomatitis, abdominal pain.

On the part of the organs of vision: conjunctivitis, dry keratoconjunctivitis.

From the respiratory system: cough, shortness of breath.

On the part of the skin and its appendages: rash, dry skin, itching.

Other: fatigue, severe infections (with or without neutropenia, pneumonia, sepsis, phlegmon).

In combination with gemcitabine for pancreatic cancer

The most common adverse events associated with Tarceva 100 mg in combination with gemcitabine, regardless of the association with the drug, are fatigue, rash, and diarrhea. Rash and grade 3–4 diarrhea occurred in 5% of patients, each requiring discontinuation of therapy in 1% of patients and dose adjustment of erlotinib in 2% of patients. The average time period before the onset of a rash is 10 days, before the onset of diarrhea is 15 days.

The combination of Tarceva 150 mg and gemcitabine increased the incidence of class-specific adverse reactions, including rash, and dose adjustment or discontinuation of therapy was required more frequently.

Hinweise

lokal fortgeschrittener oder metastasierter nicht-kleinzelliger Lungenkrebs nach Versagen einer oder mehrerer Chemotherapien;

Erstlinientherapie bei lokal fortgeschrittenem, inoperablem oder metastasiertem Bauchspeicheldrьsenkrebs in Kombination mit Gemcitabin.

Kontraindikationen

schwere Ьberempfindlichkeit gegen Erlotinib oder einen Bestandteil des Arzneimittels;

Schwangerschaft und Stillzeit.

Sorgfдltig:

Leberfunktionsstцrung;

Alter bis 18 Jahre.

Nebenwirkungen

Nebenwirkungen wurden wie folgt nach Hдufigkeit verteilt: sehr oft (=1/10); hдufig (=1/100, <1/10); selten (=1/1000, <1/100); selten (=1/10000, <1/1000); sehr selten (<1/10000), einschlieЯlich Einzelfдlle.

Monotherapie bei nicht-kleinzelligem Lungenkrebs

Die hдufigsten unerwьnschten Ereignisse, unabhдngig von der Beziehung zum Medikament: Hautausschlag (75 %) und Durchfall (54 %), von denen die meisten Grad 1 und 2 sind und keine medizinische Intervention erfordern. Hautausschlag und Durchfall 3. bis 4. Grades traten bei 9 % bzw. 6 % der Patienten mit nicht-kleinzelligem Lungenkrebs auf, die jeweils bei 1 % der Patienten einen Therapieabbruch und bei 6 % bzw. 1 % der Patienten eine Dosisanpassung von Erlotinib erforderten, bzw. Die durchschnittliche Zeitspanne bis zum Auftreten eines Hautausschlags betrдgt 8 Tage, bis zum Auftreten von Durchfall 12 Tage.

Die hдufigsten Nebenwirkungen:

Aus dem Magen-Darm-Trakt: Anorexie, Durchfall, Ьbelkeit, Erbrechen, Stomatitis, Bauchschmerzen.

Seitens der Sehorgane: Bindehautentzьndung, trockene Keratokonjunktivitis.

Seitens des Atmungssystems: Husten, Atemnot.

Auf der Haut und ihren Anhдngseln: Hautausschlag, trockene Haut, Juckreiz.

Sonstiges: Mьdigkeit, schwere Infektionen (mit oder ohne Neutropenie, Lungenentzьndung, Sepsis, Phlegmone).

In Kombination mit Gemcitabin bei Bauchspeicheldrьsenkrebs

Die hдufigsten unerwьnschten Ereignisse im Zusammenhang mit Tarceva 100 mg in Kombination mit Gemcitabin, unabhдngig von der Assoziation mit dem Arzneimittel, sind Mьdigkeit, Hautausschlag und Durchfall. Hautausschlag und Durchfall 3. bis 4. Grades traten bei 5 % der Patienten auf, die jeweils bei 1 % der Patienten einen Abbruch der Therapie und bei 2 % der Patienten eine Dosisanpassung von Erlotinib erforderten. Die durchschnittliche Zeitspanne bis zum Auftreten eines Hautausschlags betrдgt 10 Tage, bis zum Auftreten von Durchfall 15 Tage.

Die Kombination von Tarceva 150 mg und Gemcitabin erhцhte die Inzidenz klassenspezifischer Nebenwirkungen, einschlieЯlich Hautausschlag, und eine Dosisanpassung oder ein Absetzen der Therapie war hдufiger erforderlich.
