Hematopoietic growth factor.

Pegfilgrastim is a covalent conjugate of filgrastim, a recombinant human granulocyte colony stimulating factor (G-CSF), with one molecule of 20 kDa polyethylene glycol (PEG), with a prolonged action as a result of a decrease in renal clearance. Similar to filgrastim, pegfilgrastim regulates the formation and release of neutrophils from the bone marrow, markedly increases the number of neutrophils with normal or increased functional activity (chemotaxis and phagocytosis) in the peripheral blood within 24 hours and causes a slight increase in the number of monocytes and / or lymphocytes.

Like other hematopoietic growth factors, G-CSF can stimulate endothelial cells in vitro.

A transient increase in white blood cells (leukocytosis) is an expected consequence of pegfilgrastim therapy. corresponds to its pharmacodynamic effects. No side effects directly related to such leukocytosis have been described.

Single administration of pegfilgrastim after each cycle of myelosuppressive cytostatic therapy reduces the duration of neutropenia and the incidence of febrile neutropenia similarly to daily administration of filgrastim

Pegfilgrastim ist ein kovalentes Konjugat von Filgrastim, einem rekombinanten humanen Granulozyten-Kolonie-stimulierenden Faktor (G-CSF), mit einem 20-kDa-Polyethylenglycol (PEG)-Molekül mit einer verlängerten Wirkung infolge einer verringerten renalen Clearance. Ähnlich wie Filgrastim reguliert Pegfilgrastim die Bildung und Freisetzung von Neutrophilen aus dem Knochenmark, erhöht deutlich die Anzahl von Neutrophilen mit normaler oder erhöhter funktioneller Aktivität (Chemotaxis und Phagozytose) im peripheren Blut innerhalb von 24 Stunden und bewirkt eine leichte Erhöhung der Anzahl von Neutrophilen Monozyten und/oder Lymphozyten.

Wie andere hämatopoetische Wachstumsfaktoren kann G-CSF Endothelzellen in vitro stimulieren.

Ein vorübergehender Anstieg der weißen Blutkörperchen (Leukozytose) ist eine erwartete Folge der Pegfilgrastim-Therapie. entspricht seinen pharmakodynamischen Wirkungen. Es wurden keine Nebenwirkungen beschrieben, die in direktem Zusammenhang mit einer solchen Leukozytose stehen.

Die einmalige Gabe von Pegfilgrastim nach jedem Zyklus einer myelosuppressiven zytostatischen Therapie reduziert die Dauer der Neutropenie und die Inzidenz einer febrilen Neutropenie ähnlich wie die tägliche Gabe von Filgrastim
